dc.description.abstract | ในการศึกษานี้คาดว่าอนุพันธุ์โครโมนจะสามารถยับยั้งเอนไซม์แอลฟ่ากลูโคซิเดสได้ อย่างมีศักยภาพ การสังเคราะห์สารบางตัวทำเพื่อเพิ่มปริมาณให้เพียงพอสำหรับการทดสอบฤทธิ์ทางชีวภาพเท่านั้น การทดสอบฤทธิ์ยับยั้งเอนไซม์แอลฟ่ากลูโคซิเดสของอนุพันธ์โครโมนได้ทดสอบสารทั้งสิ้น 41 ตัว พบว่ามีสาร 16 ตัว (สารหมายเลข 19-20, 23-24, 32-38, 40 และ 42-45) มีศักยภาพที่ในการยับยั้งเอนไซม์แอลฟ่ากลูโคซิเดสสูงกว่าอะคาร์โบสที่มีการค่าการยับยั้ง 20.39% อนุพันธ์ที่ไม่มีหมู่แทนที่ ที่ตำแหน่งที่ 3 ตัวมีค่าการยับยั้งอยู่ในช่วง 2.38-67.85% อนุพันธ์ที่มีหมู่แทนที่ ที่ตำแหน่งที่ 3 ส่วนใหญ่แสดงการยับยั้ง > 16.57% และการยับยั้งสูงสุดมีค่า 69.24% (ยกเว้นสารหมายเลข 39 และ 46-48) อนุพันธุ์โครโมนที่มีศักยภาพมากที่สุด 16 ตัว (สารหมายเลข 19-20, 23-24, 32-38, 40 และ 42-45) และอะคาร์โบสถูกเลือกมาทำการทดสอบเพื่อหาค่า IC50ซึ่งพบว่าสารหมายเลข 33 และ 24 เป็นสารที่ยับยั้งเอนไซม์แอลฟ่ากลูโคซิเดสที่มีศักยภาพมากที่สุด 2ลำดับ โดยมีค่า IC50 เท่ากับ 5.06 และ 12.42 ไมโครโมลาร์ ตามลำดับ เพื่อให้ทราบถึงการจับกันระหว่างอนุพันธุ์โครโมนและเอนไซม์แอลฟ่ากลูโคซิเดส จึงทำการศึกษาโมเลกุลาร์ด๊อกกิงของอนุพันธุ์โครโมนที่ออกฤทธิ์ยับยั้งเอนไซม์ได้ 25 ตัว โดยใช้โปรแกรม AutoDock 4.2 ซึ่งพบว่าผลการศึกษาโมเลกุลาร์ด๊อกกิงสอดคล้องกับข้อมูลการยับยั้งแอลฟ่ากลูโคซิเดสที่ได้จากการทดลอง | th |
dc.description.abstract | In this study, chromone compounds are expected to act as potential α-glucosidase inhibitors. The synthesis was performed to increase the yield of some compounds to get sufficient yield for the biological activity experimental. The ability of forty-one chromone derivatives to inhibit α-glucosidase was determined. Among these compounds, sixteen compounds (19-20, 23-24, 32-38, 40 and 42-45) showed higher potency than acarbose with 20.39% inhibition. The 3-unsubstituted derivatives showed 2.38-67.85% inhibition. Most of the 3-substituted chromone compounds displayed > 16.57% inhibition and up to 69.24% inhibition (except compounds 39 and 46-48). Sixteen most potent chromone compounds (Compounds 19-20, 23-24, 32-38, 40 and 42-45) and acarbose were selected to determine the IC50 values. Compounds 33 and 24 were the two most potent α-glucosidase inhibitors with IC50 values of 5.06 and 12.42 µM, respectively.
