Curcumin is the major bioactive compound derived from Curcuma longa L. It offers several pharmacological advantages, including homeostatic, antibacterial, antiparasitic, hepatoprotective, antioxidant, and anti-inflammatory properties. Being poorly aqueous-soluble and permeable are the key factors preventing and delayed drug dissolution, both drawbacks reduce curcumin's oral bioavailability. There are numerous pharmaceutical formulations available today that have been developed to increase the solubility and bioavailability of curcumin. Numerous forms of curcumin are modified, including liposomes, nanoparticles, medicine, cocrystal formation is preferred. In order to enhance the solubility and dissolution of curcumin, the goal of this research is to create cocrystals of curcumin that contain amino acids such as histidine glutamine. tryptophan. 5-hydronytryptoohan and aspartic acid as conformers. Curcumin cocrystals were synthesized through dry and liquid-assisted grinding methods and verified for their formation by FTIR, DSC and PXRD. The new peak at wavenumbers around 3308-3661 cm and 1664-1739 spectrum represented the new intermolecular hydrogen bond of the synthesized cocrystal. The results from DSC and PXRD of the synthesized compound exhibited the change in thermogram and spectra from the parent compounds. These results supported the cocrystal formation. In addition the solubility was assessed. Most synthesized cocrystals demonstrated that liquid-assisted grinding with ethanol results in her solubility than dry grading. The best solubility cocrystal, Cur-Asp, was selected to study the dissolution profile. Compared to curcumin, the Cur-Asp which produced by liquid-assisted grinding and dry grinding methods presented a higher rate of curcumin release around 1.5 times. |
